Omeprazole (the active ingredient of Klofix) is a substituted benzimidazole belonging to a new class of antiulcer agents.
How Klofix Works?
Unlike H2 receptor blocking agents Klofix reduces acid secretion by inhibiting Hydrogen/Potassium adenosine triphosphatase, which is believed to be the proton pump of the gastric parietal cell.
This is the first time that intragastric acidity can be reduced independently of the primary stimulus as the mechanism of action of Klofix is at the terminal stage of the acid secreting process. Due to this both basal and stimulated acid secretions are controlled. Inhibition of enzyme is dose dependent and the drug has no effect on acetylcholine or histamine receptors. The onset of action of Klofix capsules is rapid and reversible control of gastric acid secretion is achieved with single daily dosage.
What is Klofix Used For?
Klofix Capsule is used for the treatment of;
- Short term treatment of duodenal ulcer
- Gastric ulcer
- Reflux Oesophagitis
- Management of Zollinger-ellison Syndrome
Before starting the treatment of gastric ulcer with Klofix, tests should be done to assure the absence of malignant gastric tumor. Moreover, this drug should not be given during pregnancy and lactation unless its use is unavoidable.
Klofix may prolong the elimination of drugs metabolized by hepatic oxidation such as diazepam, phenytoin and warfarin.
Recommended Dosage of Klofix
The recommended dosage of Klofix for Duodenal ulcer, Gastric Ulcer and Reflux Esophagitis is one capsule per day. In patients with duodenal ulcer, healing may occur within 2 weeks in most cases. Some patients may require a further two weeks treatment period. In patients with gastric ulcer and reflux esophagitis the duration of the treatment is four weeks. If the ulcer is not completely healed, the treatment may be prolonged for 6 or 8 weeks.
Dosage of Klofix in Zollinger-Ellison Syndrome
The recommended dosage is 3 capsules once daily. The dosage should be adjusted individually and treatment continued as long as is clinically indicated. More than 90% of the patients who showed poor response to other treatments have been treated effectively with one to six capsules per day. Daily doses above 4 Klofix capsules should be taken as two divided doses.
Possible Side Effects of Klofix Capsules
Klofix is generally very well tolerated. However, in some cases, the following side effects have been observed;
- Nausea
- Headache
- diarrhea, constipation and flatulence
- Skin rashes have also been observed in some patients.
Side effects of Klofix are generally mild and transient and their relation to the treatment has not been established.
